No, Prem teams not in Europe join in round 2 with the others in round 3. I think one of the relegated sides from last year's Prem joins in round 2 as well (whoever finished 18th). Regionalised for the first round but doing away with the seeding is one of the changes to the comp this year. Having VAR at the Prem grounds for ties being another.
